Accurate DNA quantification is essential in molecular biology, influencing the success of applications like PCR, sequencing, and cloning. Variability in sample purity, concentration, and integrity can impact experimental outcomes, so having a reliable measurement technique is crucial. 

The most common way to quantify DNA is using UV absorbance. This method involves measuring the absorbance/transmission of light through a liquid to determine the concentration of substances in the liquid. 

Another way to quantify DNA is to use fluorescent dyes bound to the DNA. The main distinction here is that these dyes are specific for double-strand DNA (as compared to spectrophotometric methods that measure all nucleic acids). These methods are more sensitive than UV absorbance, especially for low concentrations, and are often used to quantify DNA for next-generation sequencing.

Unlike absorbance-based methods, fluorescence-based methods require a standard curve, a set of samples with a known DNA quantity and their corresponding fluorescence. The fluorescence of the sample is compared against this curve to quantify the DNA. Although this method takes more time to set up in the lab, it is less messy, and most fluorometers will calculate the sample concentration for you.

Accurate and reproducible fluorescence-based DNA quantification requires a stable working solution. NovelBeamUSA’s dsDNA Working Solutions are extremely stable. The stability of the standards is also critical to ensuring accuracy and reproducibility, in fluorescence-based methods for DNA quantification. The NovelBeamUSA dsDNA High Sensitivity assay when used with the NovelBeamUSA FluoroVerse™ is highly selective for double-strand DNA (dsDNA) over RNA and designed to quantitate 0.5 ng-100ng of DNA in 200µL assay. Most standards are only stable for a short time. However, for the NovelBeamUSA dsDNA High Sensitivity assay, the prepared standard is stable for 15 days (see Figure 2 and Table 2).

In summary, fluorescence-based quantification provides a more sensitive and specific alternative to absorbance-based methods for DNA measurement. Although the requirement for a standard curve increases the initial preparation time, the approach ensures improved accuracy and reduced variability, particularly when using stable assays such as the NovelBeamUSA dsDNA kit in combination with the NovelBeamUSA FluoroVerse™.
